English: Preparation of native microscope slide with help of an elderberry stem (Anatomical structure of a cross section of an iris leaf - Iris germanica)
Materials: object and cover glass, tweezers, water dropper, blade, stem of elderberry, object (iris leaf). (0:00-0:27 sec.) Native, aqueous or temporary slides, as they are also called, are slides that last for a certain period of time and ordinary water is used for their preparation. They are easy and fast for preparation and not require additional reagents, as well as are quite practical for observing certain structures under a microscope. (0:28-1:00 sec.) In case the object from which the section is to be made is quite thin, an elderberry stem is used as an aid. (1:05-1:48 sec.) The procedure for their preparation begins with the preparation of the object and cover glass which should be wiped thoroughly. Two drops of water are placed on the object glass, one central large and one peripheral smaller drop of water. (1:49-3:25 sec.) The elderberry stem is cut longitudinally and a small piece of iris leaf is inserted in the middle. (3:30-3:40 sec.) The cross section is made with the help of an ordinary blade, with which the object (elderberry stem with iris leaf) is held in the right hand, and the blade in the left. At the beginning it is always necessary to catch up the object at the place where the intersection will be made. (3:41-4:20 sec.) The section is made by holding the object at eye level and moving the blade from outside to inside and with careful movements left and right, without pressing. The purpose of this procedure is to make a section as thin as possible through which light from the microscope will pass easily. (4:21-4:45 sec.) From the made cross-section, with the help of tweezers, the cross-section of the iris is taken and placed in the central drop with water on the object glass. (4:46-5:20 sec.) A cover glass is placed on it, which is lowered next to the peripheral drop of water and at an angle of 45° it moves towards the middle of the object glass until the moment when the two drops of water merge, during which it is lowered. This procedure needs to be performed precisely because otherwise air bubbles will form which will count during microscopy. (5:20-5:30 sec.) Потребен материјал: предметно и покровно стакленце, пинцета, капалка со вода, жилет, стебленце од бозел, објект (лист од перуника). (0:00-0:27 сек.) Нативните, водени или привремени препатати како што уште се нарекуваат се препарати коитраат одреден период и за нивна изработка се користи обична вода. Ова се препарати кои лесно и брзо се изработуваат, не бараат дополнителни реагенси и истите се доста практични за набљудување на одредени структури под микроскоп. (0:28-1:00 сек.) Во случај кога објектот од кој треба дасе направи пресек е доста тенок како помош се користи стебленце од бозел. (1:05-1:48 сек.) Постапката заизработка започнува со подготовка на предметно и покровно стакленце кои треба да се пребришат убаво. Врз предметното стакленце се ставаат две капки вода, една централна поголема и една периферна помала капка вода. (1:49-3:25 сек.) Стебленцето од бозел се сечи надолжно и на средина се вметнува мало парче од листот на перуника. (3:30-3:40 сек.) Напречниот пресек се прави со помошна обичен жилет, со што објектот (стебленце од бозел со лист од перуника) се држи во десната рака, а жилетот во левата. На почеток секогаш е потребно да се израмни објектот на местото каде ќе се прави пресекот. (3:41-4:20 сек.) Пресекот се прави со држење на објектот на ниво на очите и движење на жилетот од надвор кон внатре и со внимателни движења лево-десно, без да се притиска. Целта на оваа постапка е да се направи што е можно потенок пресек низ кој светлината од микроскопот ќе поминува лесно. (4:21-4:45 сек.) Од направениот пресек со помош на пинцета се зема пресекот од перуника и се става во централната капка со вода на предметното стакленце. (4:46-5:20 сек.) Врз него се става покровно стакленце кое се спушта покрај периферната капка со вода и под агол од 45° се движи кон средината на предметното стакленце се до моментот кога ќе се спојат двете капки со вода, при што се спушта. Оваа постапка потребно е да се изведе прецизно затоа што во спротивно ќе се формираат воздушни меурчина кои ќе сметаат при микроскопирањето. (5:20-5:30 сек.)
