English: Preparation of native microscope slide (Hygroscopic movements of spores of horsetail- Equisetum sp.):
Materials: object and cover glass, tweezers, water dropper, object (spores of horsetail). Horsetail spores have an additional outer covering called the perispore. The perispore is constructed of two hapters that are in the form of long strip with shovel-like extensions at the ends. They have the ability for hygroscopic movements and thus help to disperse the spores on the one hand, and protect them from external adverse influences on the other. (0:00-0:10 sec.) The procedure for making this temporary slide begins with the preparation of the object and cover glass that need to be wiped thoroughly. (0:11-1:00 sec.) Using tweezers, take from the powdery mass containing spores of horsetail and carefully sprinkle it on the object glass, place a cover glass on top at a 45° angle. (1:01-1:10 sec.) The slide prepared in this way is ready for microscopy. (1:11-1:25 sec.) Hapters have the ability for hygroscopic movements by being stretched or open in dry conditions. (1:26-2:00 sec.) Then carefully add water along the edges of the cover glass with the help of a dropper, as the water enters under the glass at the moment when the hapters come in contact with it, they bend around the spores in a split second. Потребен материјал: предметно и покровно стакленце, пинцета, капалка со вода, објект (спори од коњско опавче). Хаптерите претставуваат најнадворешниот дел од обивката на спорите кај коњското опавче, кој уште се нарекува периспор. Секоја спора има по две хаптери кои се во вид на долги ленти со лопатовидни проширувања на краевите. Тие имаат способност за хигроскопски движења и на тој начин го потпомагаат расејувањето на спорите од една страна, а од друга страна ги штитат од надворешните неповолни влијанија. (0:00-0:10 сек.) Постапката за изработка на овој привремен препарат започнува со подготовка на предметно и покровно стакленце кои треба да се пребришат убаво. (0:11-1:00 сек.) Со пинцета се зема од прашкастата маса која содржи спори со хаптери на коњско опавче и внимателно се наросува врз предметното стакленце, одозгора се става покровно стакленце под агол од 45°. (1:01-1:10 сек.) Вака подготвениот препарат е спремен за микроскопирање. (1:11-1:25 сек.) Хаптерите имаат способност за хигроскопски движења со тоа што во суви услови истите се испружени, односно отворени. (1:26-2:00 сек.) Потоа внимателно покрај рабовите на покровното стакленце се додава вода со помош на капалка, како што навлегува водата под стакленцето во моментот кога хаптерите ќе дојдат во контакт со неа истите во делот од секундата се свиткуваат околу спорите.
