English: Streak method inoculation of yeasts and molds
- Streak method inoculation of yeasts: 1. Using a previously sterilized inoculation loop and a sterile plate of MEA, a liquid culture of Saccharomyces cerevisiae is to be inoculated onto the plate. 2. The plate is to be incubated at 25 degree centigrade for 72 hours. - Streak method inoculation of molds Inoculation of samples of pure Penicillium sp. and Aspergillus niger cultures on MEA plates is to be carried out by: 1. Sterilizing a microbiological needle using an open flame until achieving a red glow. 2. After the needle has cooled, the needle is to be dipped in a test tube containing sterile 0.2% agar. 3. The needle is to be retracted carefully, careful to not touch the test tube walls or opening. 4. A plate of previously cultured Penicillium sp. or Aspergillus niger is to be opened carefully, extracting the desired inoculum with the needle from the peripheral region of the respective colonies. 5. Transfer is to be achieved onto a sterile petri dish by gently piercing the respective medium. 6. The plate is to be closed and the needle must be sterilized by an open flame or alternative equipment. 7. The plates are to be incubated at 25 degree centigrade for 5-7 days. -Постапка на засејување на квасци: 1. Со претходно стерилизирана еза, на плоча од MEA се засејува течна култура од Saccharomyces cerevisiae. 2. Плочата се инкубира на 25 °C, 72 часа. РЕЗУЛТАТ: На плочата од MEA се забележува раст на квасецот -Постапка на засејување на мувли: 1. Од чиста култура од Penicillium sp. и Aspergillus niger се засејува плоча од MEA на следниов начин. 2. Се вжарува бактериолошката игла на пламен до нејзино усвитување. 3. Откако иглата ќе се олади, се отвора епруветата со стерилен 0,2% агар, се обжарува отворот и се потопува иглата во него. 4. Внимателно се вади иглата од епруветата, без притоа да се допрат ѕидовите на истата и, откако ќе се обжари отворот, се затвора епруветата. 5. Се става епруветата на сталакот и внимателно се отвора плочата со Penicillium sp. и Aspergillus niger. 6. Со иглата, без претходно жарење, од средината на колонијата се земаат спори, така што иглата се допира до колонијата. 7. Се отвора неинокулираниот Петри сад и со иглата се прави убод во средината на плочата; 8. Садот се затвора и иглата се жари на пламен до нејзино вжарување. 9. Плочите се инкубираат на 25 °C, 5-7 дена. РЕЗУЛТАТ: На плочите од MEA се забележува раст на двете мувли. Осмислено и изведено од Софија Костандиновска, Институт за биологија, ПМФ, УКИМ, Скопје. |
