English: Schaeffer & Fulton spore staining method
Procedures: 1. Heat fixated smears are prepared from vegetative cells (12-14h) and from sporulating cells (24-30h) of Bacillus sp. 2. The smear is stained with a 5% aqueous solution of malachite green and kept under a gentle open flame for about 1 min, taking care as to not allow the applied dye to dry. 3. Excess dye is washed away with distilled water. 4. The smear is stained with safranin for 30 sec. 5. Excess dye is washed away with distilled water, and the smear is allowed to air-dry or dry using filter paper. 6. The smear is viewed using oleoimmersion. RESULTS: Vegetative cells should be dyed red (safranin), spores should be dyed green (malachite green) Planned and performed by Sofija Kostandinovska, Institute of Biology, FNSM, Ss. Постапки: 1. Се подготвува размаз од вегетативни клетки (12-14 часа) и од споруларни клетки (24-30 часа) од Bacillus sp. и се фиксира на топлина. 2. Размазот се прелива со 5% воден раствор со малахитно зелено и се држи препаратит над пламеник околу 1 минута, внимавајќи да не се исуши бојата. 3. Вишокот боја се измива со вода. 4. Размазот се прелива со сафранин за време од 30 секунди. 5. Препаратот се измива со вода и се суши со филтер хартија или пак се суши на воздух. 6. Препаратот се набљудува под олеоимерзија. РЕЗУЛТАТИ: Вегетативните клетки се обоени црвено (сафранин), додека спорите се обоени зелено (малахитно зелено).
