English: MPN (Most probable number) method:
1. Using a sterile pipette add 1 ml of water in the first five tubes containing LAPV media. 2. Add 1 ml of water, but with ten times higher dilution, in the second tube and 1 ml of water with hundred times higher dilution than the water added in the first five tubes. 3. Incubate the tubes at 37°С, 48 hours. RESULT: A positive result is considered when there is a change in the color of the medium to pink and the presence of gas in the Durham tube. 1. Се засејуваат седум епруветки со дурхамови цевчиња, со по 5 мл 5% лактоза андраде пептонска вода, така што во првите 5 се додава по 1 мл директно од примерокот. 2. Втората се засејува, исто така, со 1 мл, но со десет пати поголемо разредување, а третата со 1 мл со стократно поголемо разредување во однос на количеството засеано во првите 5 епрувети. 3. Епруветите се инкубираат на 37°С, за време од 48 часа. РЕЗУЛТАТ: За позитивен резултат се смета ситуацијата каде во епруветката дошло паралелно до промена на бојата на подлогата и продукција на гас (CO2 или H2) која се открива преку насобраниот гас во дурхамовото цевче (најмалку 1:10 од волуменот на цевчето).
